AMLOSEN-5 is a dual-action antihypertensive medication that combines the benefits of:
- Amlodipine (5 mg) – a long-acting calcium channel blocker that relaxes blood vessels, improving blood flow and reducing blood pressure.
- Atenolol (50 mg) – a cardio-selective beta-blocker that lowers heart rate and cardiac workload, helping to control hypertension and manage angina.
This combination ensures effective 24-hour blood pressure control and reduced cardiovascular risk in patients with hypertension or chronic stable angina.
Pharmacokinetics
- Amlodipine: Slowly absorbed, with peak plasma levels in 6–12 hours; half-life of 30–50 hours enables once-daily dosing.
- Atenolol: Rapid absorption with peak levels in 2–4 hours; half-life of 6–9 hours; mainly excreted unchanged in urine.
Pharmacodynamics
- Amlodipine: Reduces vascular resistance by inhibiting calcium influx into smooth muscles.
- Atenolol: Blocks β1-adrenergic receptors, slowing the heart rate and reducing myocardial oxygen demand.
Dosage
- Usual Adult Dose: One tablet daily, taken at the same time each day.
- Dose adjustments should be guided by a physician based on therapeutic response.
Contraindications
- Cardiogenic shock
- Severe bradycardia or hypotension
- Bronchial asthma or severe COPD
- Known hypersensitivity to Amlodipine, Atenolol, or any tablet component
Storage
- Store below 30°C in a cool, dry environment.
- Protect from moisture and light.
- Keep out of reach of children.
Warnings and Precautions
- Monitor blood pressure, heart rate, and signs of heart failure regularly.
- Use with caution in patients with liver or kidney impairment.
- Discontinue Atenolol gradually to avoid rebound effects such as tachycardia or hypertension.
- Not recommended during pregnancy or breastfeeding unless clearly necessary.
- Always consult a healthcare provider for personalized medical advice.
